Prime is for when the bike has been sitting for months and the carbs are completely dry. Also for when the tank has run dry. Don't park it in Prime.
On is for all times, the fuel won't flow unless the motor is kicked over or the motor is running.
There is no "Off" as the tap is vacuum controlled.
